Effect of indomethacin on the pregnant rat BABT
Damasceno,Débora Cristina; Volpato,Gustavo Tadeu; Ferrari,Cássio; Roldan,Luciana B.; Souza,Maricelma da Silva Soares.
The objective of this study was to evaluate the reproductive performance, liver morphological study and post mortem characteristics of the pregnant Wistar rats treated with indomethacin, a general COX inhibitor. Indomethacin at doses of 0 (control), 0.32, 1.68 and 8.40 mg/kg/day were orally given once daily to each group (n=10) on days 3 and 4 of pregnancy (day 0 = first day of pregnancy = positive vaginal sperm). The animals were euthanized under anesthesia on day 11 of pregnancy, and were carried out necropsy and microorganism culture study. The results showed that the doses of 0.32 and 1.68 mg/kg body weight (the therapeutic dose for humans) of indomethacin caused no embryotoxic or lethal effects. The highest dose (8.40 mg/kg) of indomethacin disturbed...

EVALUATION OF THE IMPLANTATION IN PINEALECTOMIZED AND/OR SUBMITTED TO THE CONSTANT ILLUMINATION RATS International Journal of Morphology
Coelho Teixeira,Álvaro Aguiar; Simões,Manuel de Jesus; Wanderley Teixeira,Valéria; Soares Júnior,José Maria.
In this paper we evaluated the morphologic and quantitative aspects of the implantations sites in uteruses of female pinealectomized and/or submitted to the constant illumination rats. The experimental groups were: I - normal female rats maintained in a clear/dark cycle of 12/12 hours, for two months, mated and euthanasiated in the 6th day of pregnancy (control); II - female sham-pinealectomized rats maintained in a clear/dark cycle of 12/12 hours, for two months, mated and euthanasiated in the 6th day of pregnancy (control); III - female pinealectomized rats, maintained in a clear/dark cycle of 12/12 hours, for two months, mated and euthanasiated in the 6th day of pregnancy; IV - normal female rats maintained in constant illumination for two months, mated...

Expression of Amphiregulin During the Pre- and Post-implantation Period in the Mouse Reproductive Tract OAK
LEE, Dong-Soo; YANAGIMOTO UETA, Yoshiko; SUZUKI, Hiroshi; 鈴木, 宏志.
In mammals, embryo implantation is an essential step in reproduction. Implantation is a phenomenon that involves crosstalk between the blastocyst and the maternal endometrium. However, the molecular basis of the connections between the blastocyst and endometrium is not yet fully understood. Amphiregulin is a member of the epidermal growth factor family and is known to be expressed in the luminal epithelium of the mouse uterus on 3.5 days post coitum (dpc). Thus, to clarify the mechanism of amphiregulin at fetomaternal interface, we analyzed the expression pattern of amphiregulin mRNA in the oviducts and uteri of pregnant and psuedopregnant mice by means of real-time PCR. Amphiregulin expression in the pregnant uterus dramatically increased on 2.5 dpc,...

Expression of Uterine Sensitization-Associated Gene-1 (USAG-1) in the Mouse Uterus During the Peri-Implantation Period OAK
MAEDA, Kanami; LEE, Dong-Soo; YANAGIMOTO UETA, Yoshiko; SUZUKI, Hiroshi; 鈴木, 宏志.
Rat uterine sensitization-associated gene-1 (USAG-1) mRNA is expressed in the uterus during the peri-implantation period, and its mRNA expression in uterine epithelial cells is highest on day 5 of pregnancy. On the other hand, since changes in USAG-1 mRNA expression in the mouse uterus are not seen during the estrous cycle, USAG-1 expression might be specifically regulated by embryonic factors rather than by the maternal environment. However, the expression pattern and function of USAG-1 in the mouse uterus have not been determined. Thus, we examined the tissue-specific USAG-1 mRNA expression in the uteri of ICR mice during peri-implantation using real-time quantitative PCR. Uterine tissues, such as the myometrium, luminal epithelium, and stroma, were...

Expression Patterns of the Implantation-associated Genes in the Uterus during the Estrous Cycle in Mice OAK
LEE, Dong-Soo; YANAGIMOTO UETA, Yoshiko; XUAN, Xuenan; IGARASHI, Ikuo; FUJISAKI, Kozo; SUGIMOTO, Chihiro; TOYODA, Yutaka; SUZUKI, Hiroshi; 玄, 学南; 五十嵐, 郁男; 鈴木, 宏志.
The mRNA expression patterns of EGF, HB-EGF, Amphiregulin, EGF receptor, IGF-1, CSF-1, IL-1 alpha, IL-1 beta, IL-1 receptor type 1, IL-1 receptor antagonist, LIF, COX-1, COX-2, Mucin-1, calcitonin, and rat USAG-1 mouse homologue, all of which are involved in the process of conceptus implantation to the endometrium, were examined during the estrous cycle by means of real-time quantitative PCR. COX-2, HB-EGF, LIF, Mucin-1, CSF-1, IL-1 alpha, IL-1 beta, and IL-1 receptor antagonist were temporally regulated during the estrous cycle and highly expressed during the estrous stage. In the case of COX-1, EGF, IGF-1, and EGF receptor, the highest mRNA expression was during the diestrous stage. In contrast, the rat USAG-1 mouse homologue mRNA expression did not...

Schwann cells for spinal cord repair BJMBR
Oudega,M.; Moon,L.D.F.; de Almeida Leme,R.J..
The complex nature of spinal cord injury appears to demand a multifactorial repair strategy. One of the components that will likely be included is an implant that will fill the area of lost nervous tissue and provide a growth substrate for injured axons. Here we will discuss the role of Schwann cells (SCs) in cell-based, surgical repair strategies of the injured adult spinal cord. We will review key studies that showed that intraspinal SC grafts limit injury-induced tissue loss and promote axonal regeneration and myelination, and that this response can be improved by adding neurotrophic factors or anti-inflammatory agents. These results will be compared with several other approaches to the repair of the spinal cord. A general concern with repair strategies...
